چکیده

Introduction: Cytosolic DNA of exogenous or endogenous origin triggers activation cyclic GMP-AMP synthase (cGAS), a cytosolic sensor, that activates innate immune responses through the adaptor protein STING. The latter, in turn, TBK1 and IKK kinases that, activate IRF3 NF-κB transcription factors, which induce expression interferons (IFNs), chemokines cytokines involved anti-tumor responses. patterns STING Hodgkin Reed-Sternberg (HRS) cells potential role cGAS-STING pathway classical lymphoma (cHL) remain unknown to date. Methods: In this prospective study, was immunohistochemically analysed cohort 52 previously untreated patients with cHL available tissue for flow cytometry (FC) analysis histology as well peripheral blood samples FC. An arbitrary 10% cutoff used positivity HRS. vitro system included 6 cell lines (MDAV, L1236, L428, L540, HDLM2, KMH2). Gene (mRNA level) expression/activation components at baseline experimental conditions were by quantitative RT-PCR (RT-qPCR) Western blot, respectively. treated agonist subjected silencing gene using transient transfection specific siRNA construct. cGAS-STING-associated IFNs cytokines, including IFN-β, CXCL10, IFN-γ, STING, control (GAPDH), RT-qPCR. Results: positive HRS 22 (42%) patients. significantly correlated lower numbers STING+ T-lymphocytes (p < 0.0001) CD3+ = 0.04) but higher CD20+ B-cells assessed FC tissues. presentation, T-cells 0.005) NK-cells 0.02). did not correlate clinical features age, gender, Ann Arbor stage, B-symptoms, tumor burden, bulky disease, anemia, other hematologic values. mRNA level substantially L428 HDLM2 compared lines. Treatment agonists stimulated IFN-β and/or CXCL10 variable indicating functional Knocking down resulted dramatic increase cells. Conclusions: is expressed subset correlates lymphocytic populations microenvironment blood. suggesting may have therapeutic implications cHL. The PRDX2 gene is transcriptionally silenced and de novo methylated in Hodgkin and Reed-Sternberg cells of classical Hodgkin lymphoma.

In a recent issue of Blood, Agrawal-Singh et al reported the identification of PRDX2 (peroxiredoxin 2) as a novel tumor suppressor gene in acute myeloid leukemia (AML). The authors observed recurrent histone H3 deacetylation and DNA hypermethylation at the PRDX2 promoter region in primary leukemia samples. Epigenetic alterations correlated with low expression of PRDX2 protein and were associate...

CD20 Over-Expression in Hodgkin-Reed-Sternberg Cells of Classical Hodgkin Lymphoma: the Neglected Quest

We have scrutinized a previously analyzed cohort of classical Hodgkin lymphoma patients for evidence of a CD20 over-expression. This was pursued in order to determine whether all the 24 (12.6%) CD20+++ patients had clinical and/or biological profiles which would warrant a separate consideration and treatment or would carry a different outcome from our 166 CD20 (-) classical Hodgkin lymphoma pat...

Tumor-infiltrated immune response correlates with alterations in the apoptotic and cell cycle pathways in Hodgkin and Reed-Sternberg cells.

PURPOSE To analyze tumor-microenvironment relationships in Hodgkin lymphoma (HL) as potential determinants in the decision-making process related to the alterations in cell cycle and apoptotic pathways of Hodgkin/Reed-Sternberg (H/RS) cells. EXPERIMENTAL DESIGN Based on a cohort of 257 classic HL patients, we carried out a global descriptive correlational analysis and logistic regression stud...

CASE STUDY Hodgkin and Reed-Sternberg cells in bone marrow aspirations of a patient with advanced classical Hodgkin lymphoma

INTRODUCTION Hodgkin Lymphoma (HL) is a group of lymphomas of mainly nodal origin with similar clinical and histological features (1). HL encompasses 95% of classical Hodgkin lymphomas (CHL) and the rest are of nodular lymphocyte predominant Hodgkin lymphoma (NLPHL). These two entities differ in their aetiology, clinical features, histopathological features, immunophenotype and molecular geneti...

Interleukin 13 and interleukin 13 receptor are frequently expressed by Hodgkin and Reed-Sternberg cells of Hodgkin lymphoma.

Hodgkin lymphoma (HL) is characterized by the abnormal expression of multiple cytokines, accounting for its unique clinicopathologic features. We have previously shown that interleukin-13 (IL-13) is secreted by HL cell lines and may serve as an autocrine growth factor. To determine the frequency of IL-13 expression in lymphoma patients, tissue sections from 36 patients with classical HL, 5 pati...
